About the Author

Dr. Terry Kenakin is Professor of Pharmacology at the University of North Carolina School of Medicine. Prior to this, he spent 7 years in drug discovery at Burroughs-Wellcome. He then moved to GlaxoSmithKline for 25 years. Dr. Kenakin has written 11 books on Pharmacology, is the Editor in Chief of the Journal of Receptors and Signal Transduction, is on numerous Editorial Boards. He is the Editor-in-Chief of Comprehensive Pharmacology (Elsevier, 2022). He is the recipient of the 2008 Poulsson Medal for Pharmacology awarded by the Norwegian Society of Pharmacology for achievements in basic and clinical pharmacology and toxicology. He has also been awarded the 2011 Ariens Award from the Dutch Pharmacological Society and the 2014 Gaddum Memorial Award from the British Pharmacological Society, and the 2020 Goodman and Gilman Award in Receptor Pharmacology from ASPET.

Description: This is an update of a compendium of the tools and methods used to quantify drug actions in biological systems and a broad description of topics relevant to drug discovery and design. The first edition was published a decade ago with revisions in 2006 and 2008.
Purpose: This book emerged from a series of courses the author taught at GlaxoSmithKline on drug action intended to bridge the communication gap between chemists and pharmacologists in assessing the actions of drugs in complex biological systems, where chemists frequently struggled to grasp the dynamics of system variability in biological models. Through revisions over the years, the author has sought to, and succeeds in, capturing emerging concepts in quantitative pharmacology and putting them in context alongside the core principles of drug-receptor interaction analysis. The book succeeds in providing a detailed, unified presentation of the concepts that drive drug design.
Audience: It was initially targeted at biologists not specifically trained in pharmacology. It addresses the principles of quantitative pharmacology at a level consistent with that of a practicing professional or interdisciplinary team of researchers. The details on the background behind the principles, their derivation, and application are sufficient for pharmacologists at any level, and the chapters on the history of pharmacology as a discipline, experimental design, and drug discovery strategies are an excellent source of rarely-addressed introductory material for new students in the field. The author is internationally recognized as a preeminent expert in the discipline who has contributed extensively to our knowledge and modeling of drug-receptor interactions.
Features: The book addresses areas such a drug-receptor theory, drug discovery, drug structure, screening strategies, pharmacokinetics and safety pharmacology, statistics, and experimental design. Introductory chapters on the history of pharmacology are a particularly rich addition. Throughout the book, the outstanding use of high-quality, data-driven figures and practical examples of relevant experiments adds to its impact. A meaningful glossary and adequately detailed index are included as well.
Assessment: Calling this a primer is something of a misnomer. It is a detailed compendium of the tools and methods used to quantify drug actions. It expands significantly on the author's now-classic Pharmacologic Analysis of Drug-Receptor Interaction, 3rd edition (Lippincott Williams & Wilkins, 1997), by transforming that small, dense treatise on quantitative analysis of drug binding into a much broader but still detailed description of the physicochemical phenomena that underlie the actions of drugs on biological systems. This edition adds solid overviews of emerging topics in the field, such as biased signaling and allosteric interactions, which keep it at the cutting edge of receptor pharmacology. Overall, this is a valuable resource for students of pharmacology and all members of drug discovery and design teams.
