Arthritis Research: Methods and Prools is a compendium of data pertinent to the methods and prools that have contributed to recent advances in molecular medicine in general, but to the molecular basis of rheumatic disease in particular. Description: This second volume in a two-volume set describing techniques used in the analysis of arthritis pathophysiology focuses on immunobiology, animal models of arthritis, and relevant new technologies useful in arthritis research.
Purpose: This volume focuses on the relevant techniques applicable to the analysis of tissues involved in arthritic disease, specifically relating to immunobiology, animal models, and emerging technologies. This volume helps researchers understand why and how a variety of investigative modalities are used.
Audience: The editor has targeted this series at clinicians and researchers in both the clinical and basic science realm who investigate the pathophysiology of arthritic disease. While there are a few chapters that may be immediately applicable for clinicians, the majority of the book focuses on technologies that exist largely within a basic science laboratory. Dr. Cope is well published in the area of the role of TNF and T cells in autoimmune and chronic inflammatory states. He is on the faculty at the Kennedy Institute of Rheumatology in London, a premier research center that first investigated the role of anti-TNF agents in rheumatoid arthritis.
Features: The three sections in this volume cover immunobiology, animal models of arthritis, and application of novel technologies in the investigation of arthritis pathophysiology. The immunobiology section comprehensively covers the methods of detection, phenotypes, and function of immunologically relevant cell types, including B and T cells, natural killer cells, and dendritic cells. This is the strongest section of the two volumes, with succinct and well described protocols for the multitude of techniques described. The second section presents the major animal models of arthritis and the evaluation of disease severity. The models include both rat and mice species and focus on the induction of inflammatory arthritis; a chapter is also included on a murine model of osteoarthritis. Particular attention is paid to the balance between the simplicity and consistency of each animal model and the inconsistencies inherent in the model to chronic human arthritic pathology. Finally, the last section delves into newer technologies that have been applied to rheumatologic diseases, from RNA analysis (including microarrays and differential display RT-PCR), protein characterization, phage display selection, and targeting of intracellular pathways using adenovirus gene delivery systems. Because this section focuses on the discovery of possible therapeutic targets, there is no unifying technical theme, unlike the first two sections. Informative figures outline all techniques, including several color plates located in the middle of the book.
Assessment: This volume excels in the presentation of basic, fundamental techniques requisite in the analysis of arthritic disease. Especially well done are the immunobiology and animal model sections. What separates this series from other references in its category is its brevity but thoroughness in the discussion of technically challenging methods.
