Description: This is an edited collection of review articles describing recent advances and current practices in cancer risk assessment. The book describes various tests of genetic toxicology, current regulatory guidelines, models of multistage carcinogenesis, and how these various subjects influence quantitative risk assessment of both genotoxic and nongenotoxic carcinogens.
Purpose: The editor's goals to provide a comprehensive overview of the practice of regulatory genetic toxicology and cancer risk assessment and to bridge the gap of communication among the spectrum of scientists engaged in different aspects of these fields generally are met. These goals are worthwhile and pertinent given the recent advances in risk assessment methodologies and the refinements and validation of genetic toxicology tests.
Audience: Although the editor and the individual contributors are acknowledged experts in their respective fields, the book suffers from a lack of clearly defined audience. Based on the stated purpose in the preface, the assumed target audience is scientists engaged in the various aspects of quantitative cancer risk assessment. However, individual chapters vary significantly in the level of detail that is presented, ranging from fairly brief overviews to a detailed protocol (chapter 7, "The mouse lymphoma assay"). The different styles are distracting and suggest perhaps the lack of a defined audience. The book certainly will be useful for graduate students interested in genetic toxicology.
Features: The book is organized into four sections plus an introduction (chapters 1-3) that provides background. Chapters 4-10 describe various methodologies for the identification of genotoxic hazards and include two chapters describing current regulatory guidelines for the interpretation and application of genotoxicity data. The second section (chapter 11) discusses quantitative aspects of dose-response relationships whereas the third section (chapter 12) concerns the use of intermediate biomarkers and molecular epidemiology to assess human exposures to potential carcinogens. The final section (chapters 13-15) deals with the characterization of risk based on the approaches outlined earlier. The organization of the book is logical, with each chapter building toward the final section. Although well organized, the individual submissions are not uniformly excellent. The book is engaging and informative mostly in the last sections, chapters 11-15. For example, the chapters on risk assessment of genotoxic and non-genotoxic carcinogens are well organized, provide excellent case studies as examples, and highlight the regulatory process efficiently and effectively. In contrast, the introductory chapter on human cancer genetics is dense with particularly unreadable figures. Also, chapters 2 and 5 covering genetic toxicology tests seem largely redundant. Similarly, chapters 9 and 10 discussing regulatory guidelines from different perspectives (agencies) perhaps could have been condensed into one chapter comparing different sets of guidelines. It also is not clear why the mouse lymphoma and micronucleus assays are singled out for full chapter descriptions, whereas it is emphasized that the salmonella (Ames) test best predicts rodent carcinogenicity.
Assessment: Overall, this book is a useful contribution to the field of genetic toxicology. It does fill a niche not currently occupied by any similar book. The emphasis on regulatory processes and the descriptive methodologies go beyond what is currently available in other more general toxicology books and as such will be useful for graduate students. Although the current practices in cancer risk assessment are well presented, the book missed an opportunity to discuss how new technologies under development (for example in genomics and proteomics) may influence future practices.
