Description: The biochemical pathways involved in the metabolism of drugs in the skin are covered in great detail in this book.
Purpose: For those who have an interest in developing topical medications or cosmetics, this should be a good resource for understanding how to create the appropriate isomers of the drug and how to predict its absorption. The writing is technical and presumes fluent understanding of biochemistry and math.
Audience: The book would be most appealing to scientists and pharmacologists who design and produce drugs for cutaneous delivery. General practitioners and dermatology students will not benefit from this book. The authors are credible authorities on the subject. Some of the authors work for the U.S. Food and Drug Administration.
Features: Mechanisms of enhancing drug delivery into the skin, nails, and follicles are discussed. Many chapters are devoted to how to quantify effects of the drug via tape stripping, blood flow assessment, and measuring the concentration of substances in the skin. I did enjoy the chapters discussing metabolism of drugs and how the cytochrome p450 system in the skin transforms compounds into a hydrophilic substance into a compound that is more easily excreted into the urine by adding polar attachments to the molecules. I also enjoyed the chapter on alternative therapies for steroids, which gave recipes for compounded preparations. I wish the author would write a book on this and discuss in more detail the rationale for mixing the ingredients and what is compatible to be mixed, cream base preferences, etc. This seems to be rapidly becoming a lost art.
Assessment: This is a well written, highly technical book that will appeal to those involved with developing pharmaceutical dermatological products. Dr. Maibach has been the editor of numerous similar texts.
